The fire was reported just after 1pm and Barrack Street was closed to traffic.
Emergency crews responded to a fire at an adult shop in Perth's CBD on Wednesday afternoon.
St John Ambulance said a man was taken to Royal Perth Hospital with burns and smoke inhalation but he has since died.
Crowds gathered to watch firefighters battle the blaze which caused significant damage to the buildings including the Club X adult shop.
